Highway 401 er en motorvei i den canadiske provinsen Ontario.
Den løper 828 km fra Windsor i vest til grensen mot Quebec i øst. Den delen av Highway 401 som passerer gjennom Toronto er Nord-Amerikas mest trafikkerte motorvei,[1][2] og en av de bredeste.[3][4]
